How do I “enable the USB drive for disk use?

I assume it's a USB hard drive, not a flash drive...so you need to partition and format the drive. Plug it in, go to Start>Control Panel>Administrative Tools>Computer Management. In the explorer part, under Storage, click Disk Management. Then find the drive, partition it, and format it.
